DTT assays

DTT (Dithiothreitol) is a common reducing agent in biochemistry.  Removal of DTT is required for Cys-based protein labeling and for disulfide bond formation in proteins.  ProFoldin offers two DTT assays to avoid interference by other molecules. Both assays measures micromolar concentrations of DTT.

The Colorimetric DTT Assay Kit (Catalog No. DTT100C) measures light absorbance at 590 nm. It is based on the strong reducing property of DTT. The assay is not interfered by phosphate buffer, mono thio compounds such as cysteine or chelators. It is interfered by NADH or NADPH.

The MicroMolar DTT Assay Kit (Catalog No.  DTT200) measures fluorescence at 535 nm with excitation at 485 nm. It is based on the chelating property of DTT. The assay is interfered by phosphate buffer, thio compounds such as cysteine, histidine and other chelators.

The Colorimetric DTT Assay Kit (Catalog No. DTT100C) includes 1 ml of 10 x Buffer, 0.2 ml of 10 x Reagent A, 0.2 ml of 10 x Reagent B and 1 ml of 10 x Reagent C.  It is for measurement of 100 samples using 96-well plates.  Cuvettes may also be used for measurements. 

The MicroMolar DTT Assay Kit (Catalog No. DTT200) includes 5 ml of Dye C55.  It is for measurement of 200 samples using 96-well plates.  Cuvettes may also be used for measurements.
